There are many public bus companies operating the Lima to Nazca route. The only problem is the headache you will have trying to compare prices and after you choose one, having to find the company’s terminal since there is no central bus terminal.

If I am in Huacachina and want to go to Arequipa, can I still do the tour in one day?

Indeed you can. Taking the private shuttle from Huacachina allows you to get early in Nazca, take the Cahuachi Pyramid tour at 3:00 pm and then get on the Peru Hop bus to continue your journey to Arequipa. If you want to take the 8:00 am tour, at least one night in Nazca is mandatory.
