If you arrive on the 22.00pm bus from Cusco, then you can do the tour if going to Arequipa in the night. If you are going to Bolivia, then you will need to spend the night.

If you are arriving from Arequipa, then you will also have time to do the tour. If you are arriving from Bolivia, then you will need to spend the night and take the night bus departing Puno the following day.
